Pepstatin is a potent inhibitor of aspartyl proteases. It is a hexa-peptide containing the unusual amino acid statine, having the sequence Isovaleryl-Val-Val-Sta-Ala-Sta. It was originally isolated from cultures of various species of Actinomyces due to its ability to inhibit pepsin at picomolar concentrations. |

Pepstatin is a peptide inhibitor isolated from cultures of Actinomycestes by Umezawa et al. (1970) and is widely used as a potent inhibitor of cathepsin E. Because this inhibitor is a general inhibitor of aspartic peptidases of the A1 family, however, it is not specific for cathepsin E. |
It is suggested that the statyl residue is responsible for the unusual inhibitory capability of pepstatin and that statine is an analog of the previously proposed transition state for catalysis by pepsin and other acid proteases.
